Abstract:
     The present invention concerns an improved process for preparation of 4'-deoxy doxorubucin from 4'-epi-N-trifluo roacetyl donorubucin as starting material. The latter is dissolved in anhydrous methylene chloride, the resulting solution is reacted with trifluoromethyl sulfonic anhydride in the presence of pyridine to obtain 4'-epi-4-trifluoromethylsulfonyloxy-N-trifluoroacetyl doxorubucin, which is reacted with tetrabutyl ammonium iodide to get 4'-deoxy-4'-iodo-N-trifluoroacetyl donorubucin, and then a reduction dehalogenation reaction is carried out by using tributyltin hydride to give 4'-deoxy donorubucin which is treated by bromine to get the corresponding 14-bromo derivative. This latter derivative is finally hydrolyzed aqueous solution of sodium formate to produce 4'-deoxydoxorubucin which is seperated as its corresponding hydrochloride form.
